Description: Hu-antigen R (HuR), also know as ELAVL1, is a ubiquitously expressed member of the embryonic lethal abnormal vision (ELAV) family of RNA-binding proteins. Three other closely related neural proteins, called HelN1 (or HuB), HuC and HuD, are target antigens in paraneoplastic neuropathy. HuR is relatively low abundance, about 5x10e5 molecules per HeLa cell. Overexpression of HuR in transiently transfected mammalian cells leads to stabilization of reporter mRNAs carrying AU-rich elements (AREs) in their 3' untranslated regions. Although immunofluorescence studies show HuR to be predominantly nuclear in interphase cells, it has been demonstrated to shuttle between the nucleus and cytoplasm. Its shuttling signal, HNS, located in the hinge region between its second and third RRMs, appears similar but not identical to the M9 shuttling sequence first identified in hnRNP A1. It has been suggested that HuR may initially bind ARE-containing mRNAs in the nucleus and accompany them to the cytoplasm, providing ongoing protection from the decay machinery. A recent report indicates that overexpression of HuR in ovarian carcinoma is associated with increased COX-2 expression, and is a prognostic factor. Applications: Suitable for use in Western Blot, ELISA, Immunoprecipitation, Immunofluorescence and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1:20-1:100 ELISA: 1:100-1:1000 Immunoprecipitation (Native): 1:10-1:20 Immunofluorescence: 1:20-1:100 Immunohistochemistry (Paraffin): 1:100-1:1000. Requires heat induced epitope retrieval (HIER) using citrate buffer in a pressure cooker for 5 minutes (1). Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
