Inducible Nitric Oxide Synthase (iNOS, iNOS 2, mNOS)
Description:
Nitric oxide (NO), a diffusible free radical gas, acts as a neurotransmitter in brain and peripheral nervous system. It accounts for the activity of endothelium-derived relaxing factors, which stimulate vasodilatation by releasing NO from the endothelium. Unlike typical neurotransmitter, NO is a not stored in synaptic vesicle and does not act on membrane receptors. Synthesis of NO, initially demonstrated in vascular endothelium, is now found in many tissues.

Applications:
Suitable for use in Immunofluorescence, ELISA, Western Blot and Immunocytochemistry. Other applications not tested.

Recommended Dilutions:
Immunofluorescence: 1:500
Western Blot: 1:1000 detects at band at ~130kD
Optimal dilutions to be determined by the researcher.

Storage and Stability:
May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Specificity:
Recognizes human iNOS (985-1002) and recombinant human iNOS (Type II). Has ~80% crossreactivity with human eNOS (1017-1034) and recombinant human eNOS (Type III). Does not react to human nNOS (1256-1273) and recombinant human nNOS (Type I). It has been epitope mapped using 96 overlapping 18 amino acid long synthetic peptides which cover the entire 1153 amino acid length of hiNOS and was found to bind to a defined region of the hiNOS sequence, residues 985-1002.
