One of the type I interferons produced by fibroblasts in response to stimulation by live or inactivated virus or by double-stranded RNA. It is a cytokine with antiviral, antiproliferative, and immunomodulating activity. The synthesis of IFN-beta can be induced by common inducers of interferons, including viruses, double-stranded RNA, and micro-organisms. It is induced also by some cytokines such as TNF and IL-1 . In contrast to IFN-alpha, IFN-beta is strictly species-specific. IFN-beta of other species is inactive in human cells. IFN-beta is involved in the regulation of unspecific humoral immune responses and immune responses against viral infections. IFN-beta increases the expression of HLA class I antigens and blocks the expression of HLA class II antigens stimulated by IFN-gamma. Recombinant Human Interferon-Beta 1a is a single, glycosylated polypeptide chain containing one N-glycosylation site, 166 amino acids and having a molecular mass of ~23kD Sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Ser-Tyr-Asn-Leu-Leu. Accession Number: V00534 Specific Activity: ~1x10e8u/mg Biological Activity: Interferon Beta-1a is fully biologically active when compared to standard. The specific activity as determined in the cytopathic effect inhibition assay as described [Rubinstein, et al.. In this antiviral assay for interferon about 1u/ml of interferon is the quantity necessary to produce a cytopathic effect of 50%. The units are determined with respect to the international reference standard for human interferon beta (Hu-IFN-b) provided by the National Institutes of Health. Unit of activity was measured on VERO cells with vesicular stomatitis virus (VSV). Storage and Stability: Aliquot to avoid repeated freezing and thawing and freeze at -70°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Aliquots are stable for at least 12 months. Molecular Weight: ~23kD |
