The major interferon produced by mitogenically or antigenically stimulated lymphocytes. It is structurally different from type I interferon and its major activity is immunoregulation. It has been implicated in the expression of class II histocompatibility antigens in cells that do not normally produce them, leading to autoimmune disease. Interferon gamma is produced mainly by T-cells and natural killer cells activated by antigens, mitogens, or alloantigens. It is produced by lymphocytes expressing the surface antigens CD4 and CD8. The synthesis of IFN-gamma is induced, among other things, by Interleukin-2 , FGF-basic , and EGF .

Recombinant Porcine IFN-gamma is a single, non-glycosylated, polypeptide chain containing 146 amino acids and having a molecular mass of 17.14kD.

Amino Acid Sequence:
The sequence of the first five N-terminal amino acids was determined and was found to be Ser-Tyr-Cys-Gln-Ala.

Biological Activity:
Porcine IFN-g is fully biologically active when compared to standard. The ED50 as determined by the amount of interferon that inhibited 50% of the cytopathic effect of vesicular stomatitis virus in MDBK cells was < 0.3 ng/ml.

Protein Content:
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm using the absorbency value of 0.556 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a calibrated solution of IFN-g as a Reference Standard.

Storage and Stability:

Lyophilized powder may be stored at -20C. Reconstitute to nominal volume by adding sterile dH2O, 0.1% BSA or HSA. Aliquot and store at -20C. Reconstituted product is stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
