Description: KSR was initially considered to be a protein kinase acting in the Ras pathway, but has more recently been demonstrated to act as a scaffold protein, facilitating complex formation between RAF and MEK and ERK. Applications: Suitable for use in ELISA, Immunoprecipitation and Western Blot. Other applications not tested. Recommended Dilutions: Western Blot: Endogenous levels of KSR may be too low to detect by Western Blot. Immunoprecipitation prior to Western Blot is recommended for detection of endogenous KSR. Note levels of recombinant KSR in transfected cells may be high enough to be detected by Western Blot. Immunoprecipitation/Western Blot: 4ug/ml detects KSR by Western Blot after using 10ug of this lot to immunoprecipitate KSR from 1mg of 3T3/A31 RIPA lysate. 3T3/A31 cell lysate was immunoprecipitated with 10ug of K6850 and immune complexes were resolved by electrophoresis, transferred to nitrocellulose and probed with K6850 (1:250). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system. Other bands correspond to light chain, heavy chain, and heavy chain dimer. The band above KSR may be incompletely reduced antibody, as it was present in a nonspeci c antibody control. Optimal dilutions to be determined by the researcher. Recommended secondary antibodies: I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo I1904-06H: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
