Medium for isolation and differentiation of Mycobacterium sp. Growth Analysis: E. coli ATCC 25922 Partial to complete inhibition Mycobacterium kansasii Group I ATCC 12478 Good to Excellent Mycobacterium scrofulaceum Group II ATCC 19981 Good to Excellent Mycobacterium intracellulare Group III ATCC 13950 Good to Excellent Mycobacterium fortuitum Group IV ATCC 6841Good to Excellent Mycobacterium tuberculosis H37Ra ATCC 25177 Good to Excellent Storage and Stability: Store powdered media at RT. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept at 4°C and used within a short period of time. Directions for use: 1. Suspend 37.3g in 600ml distilled water containing 12ml glycerol.2.Heat with frequent agitation and boil for one minute to completely dissolve medium. 3. Autoclave at 121°C for 15 minutes. 4. Prepare 1000ml of uniform suspension of fresh eggs under aseptic conditions. Avoid whipping air into suspension during the collection and mixing.5. Aspectially mix the 1000mlegg suspension with 600ml sterile Lowenstein medium cooled to 50-60°C, avoiding air bubbles.6. Dispense finished medium into sterile screw cap test tubes. Place the tubes in a slanted position and heat at 85°C for 45 minutes.
