Used as a medium for cultivation of Mycobacterium sp. when supplemented with glycerol and OADC. Modification of Middlebrook 7H9 or 7H11 formulations. Directions: Suspend 19.47g in 900 ml of distilled water. Add 5 ml of glycerol. Boil to dissolve the medium completely. Distribute in180 ml amounts in erlenmeyer flasks and sterilize by autoclaving at 15 lbs. pressure (121°C) for 10 minutes. Cool to 45°C and aseptically add 100 ml of Middlebrook OADC Growth Supplement. Mix well and pour into screw capped tubes. Storage and Stability: Store powdered media at RT. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept refrigerated protected from light, and used within a short period of time. Expected Cultural Response (2-4 weeks at 35-37°C): Mycobacterium smegmatis ATCC 14468 Appearance: Tan to light green, homogeneous, free-flowing powder Solubility: Pale yellow to medium amber with slight green tint, slightly opalescent with heat. pH: As Reported Endotoxin: ~10Eu/mg Gel: Gelling Important Note: This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications without the expressed written authorization of United States Biological. |
