Description:
Nonmuscle myosin class II proteins belong to the superfamily of actin-based motor proteins. They form hexamer structures composed of two ~200kD heavy chains (NMHC II) and two pairs of 20kD and 17kD light chains (MLC20 and MLC17). Nonmuscle myosins are classified according to the heavy chain isoforms; light chains are conserved. Three different isoforms that share 64-80% aa sequence identity have been detected in mammals: NMHC II-A, NMHC II-B, and NMHC II-C. Although NMHC II molecules have now been found in muscle cells as well, the name "nonmuscle myosin" was already widespread and is in use today. NMHC II-B is most abundant in aorta, brain, urinary bladder, and fetal lung compared to other NMHC isoforms. A single aa mutation in NMHC II-B in mice results in fatal defects in heart and brain, resulting in death before birth. In addition, NMHC II-B is phosphorylated by number of kinases, including casein kinase II and protein kinase C.

Applications:
Suitable for use in Western Blot. Other applications not tested.

Recommended Dilution:
Western Blot: 0.5-2ug/ml
Optimal dilutions to be determined by the researcher.

Storage and Stability:
May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
