Description:
Nebulin is a high molecular weight protein (500-800kD), localized at the N2-line of skeletal muscle myofibrils. It exhibits an elastic stretch dependence, as it maintains the same proportional distance between the z-line and the m-line as the sarcomere is stretched.

Applications:
Suitable for use in Immunofluorescence and Western Blot. Other applications not tested.

Recommended Dilution:
Immunofluorescence (IC): 1:400 shows a typical striated staining pattern of frozen sections of chicken skeletal muscle. It stains within the I-band and shows a decoration line at a distance of 0.22um from the z-line in electron micrographs.
Western Blot: 1:100-1:200 detects a band at ~500kD
Optimal dilutions to be determined by the researcher.

Storage and Stability:
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Specificity:
Recognizes chicken Nebulin. Localizes nebulin in skeletal muscle of a wide variety of species from cold-blooded vertebrates to man. Does not crossreact with smooth or cardiac muscle, non-muscle tissue and cultured cells. Localizes nebulin within the I band of skeletal muscle sections by indirect immunofluorescence. Species crossreactivity: human, mouse, rat, alligator, amphibian, avian, bovine, fish, porcine, rabbit.
