Description: Neurogranin is a 78 amino acid neuron-specific substrate of protein kinase C that accumulates predominantly in dendritic shafts and spines of neurons. Immunoreactive neurogranin has been detected in many brain regions but is particularly abundant in the mammaliam forebrain. Immunostaining is completely abolished by pre-adsorption of the antibody with the immunogen peptide. Applications: Suitable for use in Western Blot, Immunoprecipitation and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1:1000 (using ECL) Immunohistochemistry (Frozen): 1:1000 The antibody has been used successfully on paraformaldehyde-fixed cryostat or Vibratome sections. Immunoprecipitation: 1:250 on perchloric acid-treated brain homogenates using Protein A-Agarose to precipitate the immune complex. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
