Description: This is an N-ras oncogene encoding a membrane protein that shuttles between the Golgi apparatus and the plasma membrane. This shuttling is regulated through palmitoylation and depalmitoylation by the ZDHHC9-GOLGA7 complex. The encoded protein, which has intrinsic GTPase activity, is activated to a GTP-bound form by a GTPase activating protein and inactivated to a GDP-bound form by a guanine nucleotide-exchange factor. Defects in this gene are a cause of juvenile myelomonocytic leukemia (JMML). Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Western Blot: 0.01-0.03ug/ml. A band at ~21kD is seen with human lysates in A431 cell line. In transfected HEK293 transiently expressing NRAS bands of ~21kD and 25kD are seen. These bands are not seen in non-transfected HEK293. Immunohistochemistry (Paraffin): 2-3ug/ml. Shows staining of cytoplasm and membranes of enterocytes in paraffin embedded human small intestine. Optimal dilutions to be determined by the researcher. Positive Control: Human lysates of cell line A431 Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
