NRAS (GTPase NRas, Transforming Protein N-Ras, HRAS1)
Description:
This is an N-ras oncogene encoding a membrane protein that shuttles between the Golgi apparatus and the plasma membrane. This shuttling is regulated through palmitoylation and depalmitoylation by the ZDHHC9-GOLGA7 complex. The encoded protein, which has intrinsic GTPase activity, is activated to a GTP-bound form by a GTPase activating protein and inactivated to a GDP-bound form by a guanine nucleotide-exchange factor. Defects in this gene are a cause of juvenile myelomonocytic leukemia (JMML).

Applications:
Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested.

Recommended Dilutions:
Western Blot: 0.01-0.03ug/ml. A band at ~21kD is seen with human lysates in A431 cell line. In transfected HEK293 transiently expressing NRAS bands of ~21kD and 25kD are seen. These bands are not seen in non-transfected HEK293.
Immunohistochemistry (Paraffin): 2-3ug/ml. Shows staining of cytoplasm and membranes of enterocytes in paraffin embedded human small intestine.
Optimal dilutions to be determined by the researcher.

Human lysates of cell line A431

Storage and Stability:
May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
