NT-3 (Neurotrophin 3, HDNF, NGF-2, Nerve growth factor-2)
NT-3 is a neurotrophic factor structurally related to NGF, BDNF, and NT-4. These proteins belong to the cysteine-knot family of growth factors that assume stable dimeric structures. NT-3 is expressed by neurons of the central nervous systems and can signal through the trk receptors. NT-3 promotes the growth and survival of nerve and glial cells. The amino acid sequences of human, murine and rat NT-3 are identical. Recombinant human NT-3 is a noncovalently linked homodimer, of two 13.6kD polypeptide monomers (238 total amino acid residues).

Suitable for use in Neutralization, ELISA, and Western Blot. Other applications not tested.

Recommended Dilution:
Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity (50ng/ml), a concentration of 2-4ug/ml of this antibody is required.

ELISA: Detection by direct ELISA (using 100ul/well antibody solution), requires a concentration of at least 0.5ug/ml of antibody. This antigen-affinity purified antibody, in conjunction with compatible secondary reagents, allows detection of 0.2-0.4ng/well.

Western Blot: Use at a concentration of 0.1-0.2ug/ml. Used in conjunction with compatible secondary reagents the detection limit is 1.5-3.0ng/lane, under either reducing or non-reducing conditions.

Optimal dilutions to be determined by the researcher.

Storage and Stability:
Lyophilized powder may be stored at 4C for short-term only. Reconstitute to nominal volume by adding sterile 40-50% glycerol and store at -20C. Reconstituted product is stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
