Description: Expression of p21WAF1 is induced by wild type, but not mutant, p53 suppressor protein. The p21WAF1 protein binds to cyclin/CDK complexes and inhibits their kinase activity thereby stopping cell cycle progression. Meanwhile, the p21WAF1 protein also binds to PCNA (proliferating cell nuclear antigen) and blocks DNA replication but not the DNA repair process. Evidently, inactivation of p53 disrupts the p53-dependent expression of p21WAF1. p21WAF1is a tumor suppressor involved in the pathogenesis of a variety of malignancies. Applications: Suitable for use in Immunofluorescence, Immunoprecipitation and Immunohistology. Not suitable for Flow Cytometry or Western Blot. Other applications have not been tested. Recommended Dilution: Immunoprecipitation (native and denatured) : 2ug/ml protein lysate. Co-precipitates p21 proteins. Immunohistochemistry (formalin and paraffin): 1:25-1:50 for 60 minutes at RT. Staining of formalin-fixed tissues REQUIRES boiling tissue sections in 10mM citrate buffer, pH 6.0 for 10-20 min followed by cooling at RT for 20 min. Optimal dilutions to be determined by the researcher. Positive Control: Ls174T or Raji cells and colon carcinoma. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add glycerol (40-50%) and aliquot. Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
