Perforin (Cytolysin)
Description:
Perforin, originally cloned from mouse cytotoxic T-lymphocyte (CTL), is a 555-amino acid protein with a 21-amino acid signal peptide and is a potent cytolytic pore-forming protein. It is a specific marker of functionally active cytotoxic T-lymphocytes (CTLs) and natural killer (NK) cells. Fas ligand and perforin are the two key effector mechanisms in T cell-mediated cytotoxicity. These molecules mediate cytolysis of target cells by membrane damage and apoptosis, an important mechanism in the immune defense against tumors and virus infection. Perforin expression is constitutive on NK cells, but increases in resting XD8+ cytotoxic cells upon activation.

Applications:
Suitable for use in Immunoprecipitation, Flow Cytometry, Immunohistochemistry and Immunocytochemistry. Other applications not tested.

Recommended Dilution:
Immunoprecipitation: 2-8ug/ml
Flow Cytometry: 2-8ug/ml
Immunohistochemistry (frozen and paraffin sections): 2-8ug/ml
Immunocytochemistry.: 2-8ug/ml
Optimal dilutions to be determined by the researcher.

Storage and Stability:
May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
