Phosphatidylserine (PS)
Phosphatidylserine (PS) is a phospholipid found in the membrane of all plants and animals. PS is most concentrated in the brain, as PS makes up about 10% of the lipid content of neurons. Neural PS is thought to play a role in maintaining nerve cell integrity, enhancing neurotransmitter signal efficiency, and improving nerve cell signal transmission. One of the key features of apoptosis is the expresion of PS on the outer cell membrane. PS is usually found on the inner side of the plasma membrane, but during early stage apoptosis, the PS is translocated to the outer membrane. FITC-labeled annexin V can bind to the exposed PS, providing the basis of most annexin V-based apoptosis assays.

Suitable for use in Immunohistochemistry and Flow Cytometry. Other applications have not been tested.

Recommended Dilutions:
Imunohistochemistry: Reported to detect phosphatidylserine in rat carotid artery paraffin sections fixed with ethanol/acetone.
Flow Cytometry: 0.2-2ug/ml detects cell surface phosphatidylserine on Jurkat cells treated with Fas Ligand vesicles.
Dose response of Fas Ligand using surface phosphatidylserine
(PS) as readout: Jurkat cells were treated with increasing doses of Fas Ligand, Membrane Bound. After 18 hours, cells were surface stained with P4071-80, (2ug/ml) followed by an IgG F(ab)2 (PE) goat anti-mouse IgG. Percentage of cells staining positive for PS increased from ~8% for untreated cells, to 14%, 15%, and 20%, with increasing dose of Fas Ligand.
Optimal dilutions to be determined by the researcher.

Storage and Stability:
May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
