Description: This gene encodes a cGMP-binding, cGMP-specific phosphodiesterase, a member of the cyclic nucleotide phosphodiesterase family. This phosphodiesterase specifically hydrolyzes cGMP to 5'-GMP. It is involved in the regulation of intracellular concentrations of cyclic nucleotides and is important for smooth muscle relaxation in the cardiovascular system. Alternative splicing of this gene results in three transcript variants encoding distinct isoforms. Applications: Suitable for use in ELISA and Western Blot. Other applications have not been tested. Recommended Dilutions: Peptide ELISA Titer: 1:16,000 Western Blot: 0.3-1ug/ml ~100kD band observed in Human Lung lysates (calculated MW of 100kD according to isoform 1, NP_001074.2). In transfected HEK293 transiently expressing PDE5A isoform 2 a band of approx. 95kD is observed. This band is not observed in the non-transfected HEK293 (calculated MW of 94.8kD according to isoform 2, NP_236914.2). Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
