Recombinant protein expressed in yeast (S. cerevisiae) from the c-sis oncogene. Sterility: Sterilized through a 0.2um lter and packaged aseptically. Reconstitution: Dilution with 4mM HCl or 10mM acetic acid containing 1-2mg/ml BSA is recommended. Note: In dilute solution, PDGF adheres to surfaces, particularly glass. Therefore, use only polypropylene containers, pipettes and tips. Biological activity is destroyed upon reduction. Biological Activity: 5ng/ml of this lot of PDGF-BB supported a 1.9-fold increase in cell growth of NIH 3T3 broblasts over control cultures without PDGF-BB. The optimal PDGF concentration in cell culture can vary depending on cell type. PDGF Growth Assay: 1. Prepare a cell suspension in 2% platelet poor serum to a nal concentration of 1x10e4 cells/ml. Aliquot 4ml/tube to seed quadruplicate wells (1ml/well) of test, positive and negative controls. 2. Prepare PDGF-BB following instructions above for a concentration of approximately 1ug/ml and appropriate dilutions. 3. Add PDGF-BB directly to the 4ml suspension, mix well and dispense 1ml/well ( nal concentration 0-10ng/ml). 4. Refeed on day 3 with growth medium + PDGF-BB. 5. Count and record cell numbers on day 5. Calculate the stimulation factor over negative control results. Storage and Stability: Lyophilized powder may be stored at -20°C. May be stored at 4°C for short-term only. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Source: Yeast Purity: Puri ed by HPLC. SDS-PAGE Analysis: Reduced PDGF-BB runs as one band at 16-17kDa. Non-reduced PDGFBB runs as one band at 30-32kDa. Because of anomalous behavior, PDGF-BB sometimes runs as a doublet at 14-18kDa. Form: 10ug PDGF-BB, lyophilized from 40ul of 0.1M acetic acid. Quantitated by amino acid analysis.
