PDGF is a mitogenic peptide growth hormone carried in the alpha-granules of platelets. It is released when platelets adhere to traumatized tissues. Connective tissue cells near the traumatized region respond by initiating the process of replication.The synthesis of PDGF can be induced by Interleukin-1, Interleukin-6, TNF-alpha, TGF-beta and EGF.

Recombinant Human Platelet Derived Growth Factor-BB is a homodimeric, glycosylated, polypeptide chain containing 109 amino acids and having a molecular mass of 32,021 Daltons.

<1% as determined by silver-stained SDS-PAGE gel analysis.

Biological Activity:
PDGF-BB is fully biologically active when compared to standard. The ED50, calculated by the dose-dependent proliferation of murine BALB/c 3T3 indicator cells (measured by 3H-thymidine uptake) is <1ng/ml, corresponding to a Specific Activity of 1MIU/mg.

Protein Content:
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280nm using the absorbency value of 0.55 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a standard solution of PDGF-BB as a Reference Standard.

The sequence of the first five N-terminal amino acids was determined and was found to be Ser-Leu-Gly-Ser-Leu.

Storage and Stability:
Lyophilized powder may be stored at 4C for short-term only. Reconstituted product is stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Purity: 95% by RP-HPLC or reducing/non-reducing SDS-PAGE Silver Stain. Chromatographically purified.
Form: Supplied as a lyophilized powder in 20mM sodium acetate buffer, pH 7.4. Reconstitute in sterile dH2O 0.1mg/ml. Can be further diluted to other aqueous solutions.
