Placenta growth factor (PlGF), a homodimeric glycoprotein that is homologous to vascular endothelial growth factor (VEGF), is mitogenic to endothelial cells and protects trophoblast from apoptosis. Alternative splicing of mature mRNA gives rise to three known isoforms of PlGF. PlGF is expressed by human trophoblast during normal pregnancy, however, it is not known which isoforms are produced.
Recombinant Human Placenta Growth Factor-1(PIGF/PGF) produced in insect cells is a homodimer, glycosylated polypeptide chain containing 2 x 131 amino acids and having a total molecular mass of 29.7kD. Recombinant Human PLGF-1 is purified by proprietary chromatographic techniques
Amino Acid Sequence:
The sequence of the first five N-terminal amino acids was determined and was found to be Met-Leu-Pro-Ala-Val.
Dimers and Aggregates:
Less than 1% as determined by silver-stained SDS-PAGE gel analysis.
Recombinant Human PLGF-1 is fully biologically active when compared to standard. The ED50 as determined by its ability to bind to immobilized rh-sFlt-1 in a functional ELISA. Recombinant human PlGF-1 can bind to immobilized rh-sFlt-1 (100 ng/well) with a linear range at 0.5 – 10 ng/mL.
0.1ng/ug (IEU/ug) of Recombinant PLGF-1.
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm.
2. Analysis by RP-HPLC, using a standard solution of PLGF-1 as a Reference Standard.
It is recommended to reconstitute the lyophilized Recombinant PLGF-1 in sterile 20mM acetic acid not less than 100µg/ml, which can then be further diluted to other aqueous solutions.
Lyophilized Human PLGF-1 although stable at room temperature for 3 weeks, should be stored desiccated below -180C. Upon reconstitution Recombinant Human PLGF-1 should be stored at 40C between 2-7 days and for future use below -180C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please avoid freeze-thaw cycles.
