RANKL binds to tnfrsf11b/opg and to tnfrsf11a/rank. Osteoclast differentiation and activation factor. Augments the ability of dendritic cells to stimulate naive t-cell proliferation. May be an important regulator of interactions between t-cells and dendritic cells and may play a role in the regulation of the t-cell-dependent immune response. sRANKL may also play an important role in enhanced bone-resorption in humoral hypercalcemia of malignancy. Recombinant Human soluble RANKL is a single, non-glycosylated polypeptide chain containing 176 amino acids and having a molecular mass of 20006 Dalton. Sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Met-Glu-Lys-Ala-Met. Biological Activity: RANK ligand is fully biologically active when compared to standard. The activity is determined by its ability to induce osteoclast formation in RAW264.7 cells using a concentration of 5-10ng/ml and by dose dependent stimulation of IL-8 production in human PBMC. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile ddH2O, 0.1% BSA or HSA. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
