Sterility testing medium for isolation of yeasts and molds.

Sabouraud Dextrose Agar is a general-purpose medium devised by Sabouraud for the cultivation of dermatophytes. The low pH of approximately 5.6 is favorable for the growth of fungi, especially dermatophytes, and slightly inhibitory to contaminating bacteria in clinical specimens. This medium is recommended in the USP for use in performing total combined mold and yeast counts

Dissolve 30g in 800-900ml of ddH2O stirring gently until completely solubilized. Add 3.7g/L sodium bicarbonate. Adjust pH of the medium to the desired level. Add additional water to bring the solution to 1L. Heat the agar media with frequent agitation and boil for 1 minute to completely dissolve the powder. Avoid overheating. Autoclave at 121C for 15 minutes.Storage and Stability:

Store powdered media at RT. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept at 4C and used within a short period of time.

Appearance: Off-white, free-flowing, homogeneous powder.

Solubility: Solution is light amber, clear to very slightly opalescent.
