SAP102 (Synapse-associated Protein 102, Discs Large Homolog 3, DLG3, NE-DLG)
Synapse-associated protein (SAP) 102 is a 102kD membrane-associated synapse protein with extensive sequence homology to the PSD 95 family of proteins that facilitate ion channel clustering at synaptic terminal (1,2). SAP102 consists of three 90 amino acids PDZ domains at its amino terminus, followed by a Src homology (SH) 3 domain, and a region with homology to guanylate kinases (GK). SAP 102 is detected in dendritic shafts and spines of asymmetric type 1 synapses (1,2). SAP102 associates with NMDA receptor through the carboxyl terminal NR2B subunit of NMDA receptor (1,2). In addition to ion channel, it also interacts with synaptic ras-GTPase activating protein SynGAP through its PDZ domains (3). Expression of SAP 102 restores synaptic boutons in drosophila with defective Drosophila tumor suppessor protein
(DlgA), a PSD 95 related protein involved in maintaining integrity of epithelial and neuromuscular junction (4).

Suitable for use in Western Blot, Immunoprecipitation, Immunohistochemistry and Immunoelectron Microscopy. Other applications have not been tested.

Recommended Dilution:
Western Blot (Colorimetric): 4ug/ml was sufficient to detect 20ug of rat brain tissue extract by colorimetric immunoblot analysis.

Optimal dilutions to be determined by researcher.

Storage and Stability:
May be stored at 4C for short-term only. For long-term storage, store at -20C. Aliquots are stable for at least 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
