SB 203580 is a highly specific inhibitor of SAPK2a/p38 (IC50=600nM). It suppresses the activation of MAPKAP Kinase-2 and MAPKAP Kinase-3. SB 203580 does not significantly inhibit SAPK/JNK or Erk/MAP kinases at 100mM. It inhibits IL-1 and TNF-a production from LPS-stimulated human monocytes and the human monocyte cell line THP-1. SB 203580 has been shown to inhibit activation of the HIV-1 long terminal repeat by IL-1 and TNF-a. It has also been shown to be an effective inhibitor of inflammatory cytokine production in vivo in mice and rats.

Applications: Suitable for use in Protein Kinase Inhibition Assay. Other applications not tested.

Recommended Dilutions: Protein Kinase Inhibition Assay: A 20 minute pre-incubation of immunoprecipitated SAPK2a/p38 MAPK with 1-10uM of SAPK3/p38 Inhibitor (SB203580) inhibits the kinase activity 80-95%. Optimal concentration of SB 203580 needed may depend on the amount of enzyme used.

Storage and Stability:
Lyophilized powder may be stored at 4°C for short-term only. For Long term storage, store at -20°C. Lyophilized powder is stable for 36 months at -20°C. Reconstitute to nominal volume by adding sterile DMSO, aliquot and store at -20°C. Reconstituted product is stable for 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
