Description: Applications: Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Western Blot: >1:100 Immunohistochemistry: 1:500-1:1000 in PBS. Bn/SA-HRP Technique: Perfusion Fixation: Fixative-4% paraformaldehyde in 0.1M PBS, pH 7.4; 500ml over ~20 minutes. Post Fixation: 4% paraformaldehyde in 0.1M PBS, pH 7.4; 500ml over ~20 minutes. Post Fixation: 1.5 hour at 4°C. in 4% paraformaldehyde in 0.1M PBS, pH 7.4. Note: Paraformaldehyde is a necessary component in fixation of serotonin. If needed, low levels of glutaraldehyde (0.1-0.3%) may be used in conjunction with paraformaldehyde. Incubation on Tissue: 16-24 hours at 4ºC. Optimal dilutions to be determined by the researcher. Positive Control: Rat cortex and hippocampus Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
