Chemokine SDF-1 is highly conserved among species and belongs to a large family of small, structurally related cytokines. It is a chemoattractant for T lymphocytes, monocytes, and hemopoietic stem cells and is constitutively expressed in bone-marrow-derived stromal cells. Recent reports indicate that SDF-1 is the ligand for LESTR/fusin and prevents infection by T-cell-line-adapted HIV-1. SDF-1a and SDF-1b, are generated by alternative RNA splicing of transcripts encoded by a single gene. SDF-1a differs from SDF-1b by four C-terminal amino acid residues absent in SDF-1a. As yet, no functional differences have been reported. Stromal Cell-Derived Factor-1 (SDF1 ) is a recently discovered protein belonging to the -chemokine (C-X-C) family of cytokines. Human SDF1 is an 8kD protein containing 68 amino acid residues. Recombinant Human SDF-1 produced in E.Coli is a non-glycosylated, Polypeptide chain containing 68 amino acids and having a molecular mass of 8008 Dalton. The rHuSDF-1 is purified by proprietary chromatographic techniques. Amino Acid Sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Lys-Pro-Val-Ser-Leu Biological Activity: The specific activity as determined by its ability to chemoattract human peripheral T cells activated with PHA and IL-2 using a concentation of 20-80ng/ml. Protein Content: Protein quantitation was carried out by two independent methods: 1. UV spectroscopy at 280 nm using the absorbency value of 1.06 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics). 2. Analysis by RP-HPLC, using a standard solution of SDF-1 as a Reference Standard. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile ddH2O. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. Molecular Weight: 8.01kD |
