Composition per Liter:
Selective medium for Coliform organisms, based on lactose fermentation, from urine and routine water and food testing. Lactose fermenting bacteria appear as yellow colonies. Lactose non-fermenting bacteria appear as blue colonies.
Medium green, homogeneous, free flowing powder with soft lumps.
pH (1.8%, 25ºC):
Yeast Extract 3.0
Tergitol 7 0.1
Bromthymol Blue 0.25
Directions per Liter:
Dissolve 18g in 1L of ddH2O stirring gently with heating until completely solubilized. Boil for one minute to dissolve completely. Dispense into appropriate containers, loosen caps and autoclave for 15 minutes at 121ºC (15psi). After cooling medium to 45-50.C add 3.0 ml of one percent triphenyl tetrazolium chloride, if desired.
Storage and Stability:
Store powdered media at RT. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept refrigerated and used within a short period of time.
TTC Solution: Triphenyl tetrazolium chloride: 1.0g/100mlAdd Triphenyl tetrazolium chloride to ddH2O and bring volume to 100ml. Mix thoroughly and filter sterilize.
Directions per LiterDissolve 18.125 grams per liter of distilled/deionized (DDI) water, heating with stirring until completely solubilized, adjusting pH as necessary. Aseptically add 3.0ml of TTC solution. Mix thoroughly. Dispense into appropriate containers, loosen caps and autoclave for 15 minutes at 121ºC (15psi).StorageStore powdered media at RT.
