Thrombospondin (TSP)
Description:
Thrombospondin is a protein from platelet a-granules. It is made up of three identical subunits bound by interchain disulfides. It is secreted at sites of platelet activation and aggregation and is involved in the processes of chemotaxis, adhesion, proliferation and differentiation of leukocytes, fibroblasts, smooth muscle and endothelial cells.

MW of Antigen: ~450kD (Non-reduced)

Epitope: platelet-or cell-binding domain at the extreme C-terminus of TSP.

Applications:
Suitable for use in Electron Microscopy, Flow Cytometry, Immunofluorescence, Immunoprecipitation, Western Blot, and Blocking studies. Not suitable for Immunohistochemistry.
Other applications have not been tested.

Recommended Dilution:
Immunoprecipitation: 2ug/mg protein lysate (native verified; Use Protein G)
Western Blot: 1-2ug/ml for 2hrs at RT
Blocking: 25-50ug/ml blocks binding of TSP-1 to IAP/CD47 (Order Ab without azide)
Optimal dilution to be determined by the researcher.

Positive Control: HUVEC cells.

Cellular Localization: Secretory granules, Golgi complex, endoplasmic reticulum, extracellular matrix

Storage and Stability:
May be stored at 4C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50), aliquot and store at -20C. Aliquots are stable for at least 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Specificity:
Shows no crossreactivity with fibronectin, fibrinogen and von Willebrand factor.1 It blocks TSP-mediated agglutination of fixed RBCs.3 It shows no effect on TSP-mediated agglutination of fixed, activated platelets.3 It inhibits both thrombin-and A23187- induced aggregation of washed, live (not fixed) platelets without affecting the secretion of serotonin.1 It inhibits adhesion of melanoma G361 cells to TSP.4 It blocks binding of C-terminal domain to Integrin-Associated Protein (IAP) / CD47. Species Crossreactivity: human, bovine, porcine, equine, canine, sheep, rat.
