Tryptose Agar is recommended for the cultivation and isolation of pathogenic and saprophytic bacteria, especially Brucella as specified by Huddleson and Castaneda (1,2) Huddleson found Tryptose media to be equal or superior to meat infusion media, providing uniformity for the cultivation and differentiation of fastidious organisms (1).

The successful isolation of Brucella spp. supports the use of Tryptose media as a rich general purpose media. Tryptose Agar is prepared with Tryptose, which is a mixed enzymatic hydrolysate with rich nutritional value for microbiological culture media. The enzymatic hydrolysis of Tryptose permits the preservation of vitamins and amino acids.

Tryptose 20g
Dextrose 1g
Sodium Chloride 5g
Agar 15g
Total: 41g/L

Directions per Liter:
Dissolve 41g in 1L of ddH2O. Heat with frequent agitation and boil for one minute to completely dissolve the medium. Dispense into appropriate containers, loosen caps and autoclave (15psi) for 15 minutes at 121C.

Storage and Stability:
Store powdered media at RT. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept at 4C and used within a short period of time.

Certificate of Origin:
The raw animal product used in the manufacturing of Tryptose Phosphate Broth is derived from casein originating in the USA, New Zealand and/or Australia. The animals used received both ante- and post-mortem inspections under certified veterinary care and were inspected to be free of apparent infectious and contagious diseases. During processing of the raw material, a batch is heated to a minimum of 80C for 1 hour including a minimum of 5 minutes at 105C. The product is then dried at a minimum of 140C. BSE (Bovine Spongiform Encephalopathy) is not known to exist in the New Zealand and Australia.
