Differentiation of organisms based on urease production.

Mix 29g of the medium in 100mL of purified water until evenly dispersed. Sterilize by filtration. Mix 15g of agar in 900mL of purified water until evenly dispersed. Heat with repeated stirring and boil for one minute. Autoclave at 121.0C for 15 minutes. Cool sterilized agar to 45to50C and aseptically add the sterile Urea Agar Base. Mix thoroughly and dispense into sterile tubes. Place tubes in a slanted position. Storage and Stability:Store powdered media at RT. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept at 4C and used within a short period of time.

pH: 6.8 0.2

Appearance: Off-white, homogeneous, free flowing powder.

Solubility: Light to medium yellowish-orange, clear solution.
