Alternate formulation of the YPD series media, formulated with Peptone Y (casein digest, tryptone), useful for yeast propagation and plating of mutants and transformants.

Also includes the addition of an adenine hemisulfate supplement to a final 1X concentration of 120mg/L. The adenine supplement prevents yeast strains harboring ADE1 and ADE2 mutations (e.g. Y2HGold, AH109, and Y187) from turning pink/red after extended growth.

Directions per Liter: Dissolve 50 grams in 800-900ml of ddH2O stirring gently with heating until completely solubilized. Adjust pH of the medium to the desired level. Add additional water to bring the solution to 1L. Dispense into appropriate containers, loosen caps and autoclave for 15 minutes at 121ēC (15psi).

Storage and Stability: Store powdered media at RT. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept at 4°C and used within a short period of time.
