(Apoptosis regulator Bcl-2, B-cell CLL/lymphoma-2)

This antibody recognizes a protein of 25-26kDa, identified as the bcl-2alpha oncoprotein. It shows no cross-reaction with Bcl-x or Bax protein. Expression of bcl-2alpha oncoprotein inhibits the programmed cell death (apoptosis). In most follicular lymphomas, neoplastic germinal centers express high levels of bcl-2alpha protein, whereas the normal or hyperplastic germinal centers are negative. Consequently, this antibody is valuable when distinguishing between reactive and neoplastic follicular proliferation in lymph node biopsies. It may also be used in distinguishing between those follicular lymphomas that express bcl-2 protein and the small number in which the neoplastic cells are bcl-2 negative.

Applications:  Suitable for use Western Blot, Immunoprecipitation, Immunohistochemistry. Other applications not tested.
Recommended Dilution
Western Blot: 0.5-1ug/ml Immunoprecipitation: 0.5-1ug/500ug protein lysate Immunohistochemistry: Frozen & Formalin-fixed: 0.5-1ug/ml for 30 minutes at RT (Staining of formalin-fixed tissues is enhanced by boiling tissue sections in 1mM EDTA Buffer, pH 7.5-8.5, for 10-20 min followed by cooling at RT for 20 minutes Optimal dilutions to be determined by the researcher.
Positive Control
Jurkat, K562, HL-60, or HeLa Cells. Tonsil or follicular lymphomas.
Storage and Stability
Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Note: Applications are based on unconjugated antibody.
Immunogen
Synthetic peptide corresponding to aa41-54, GAAPAPGIFSSQPG-Cys, from human Bcl-2. Cellular Localization: Outer mitochondrial membranes and endoplasmic reticulum as well as nuclear membranes.
Form
Supplied as a liquid in 10mM PBS. No preservative added. BSA-Free. Labeled with horseradish peroxidase (HRP).
Purity
Purified by Protein A/G affinity chromatography.
Specificity
Recognizes human Bcl-2. Does not react with mouse and rat. Does not react with Bcl-x or Bax protein.
