MaxLight™490 is a new Blue-Green photostable dye conjugate comparable to DyLight™488, Alexa Fluor™488 and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (491nm); Emission (515nm); Extinction Coefficient 73,000.

Recognizes a 180-185kD protein, identified as isoform of leukocyte common antigen (CD45RO). The epitope recognized by this antibody is sensitive to neuraminidase digestion. This antibody reacts with mature activated T-cells, most thymocytes, and a sub-population of resting T-cells within both CD4 and CD8 subsets. It shows no reactivity with normal B or natural killer cells, but reacts with granulocytes and monocytes. Reportedly, it is useful to identify T-cell lymphomas and leukemias. It rarely stains NK cells or B-cell lymphomas.
Applications
Suitable for use in Immunofluorescence, Flow Cytometry, FLISA, Western Blot, Immunoprecipitation, Immunohistochemistry. Other applications not tested.
Recommended Dilution
Flow Cytometry: 0.5-1ug/million cells Immunofluorescence: 1-2ug/ml Western Blot: 0.5-1ug/ml Immunoprecipitation: 1-2ug/500ug protein lysate Immunohistochemistry (Frozen & Formalin-fixed): 0.5-1ug/ml for 30 minutes at RT (Staining of formalin-fixed tissues requires boiling tissue sections in 10mM Citrate Buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 minutes) Optimal dilution for a specific application should be determined.
Positive Control
CCRF-CEM, Jurkat or MOLT-4 cells. Paracortex in a tonsil or a reactive lymph node.
Storage and Stability
Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™490 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. 
Note: Applications are based on unconjugated antibody.
