Rat Anti-TIM-4 (T-cell Immunoglobulin Mucin 4, Tim4, Timd4) (FITC)

Tim-4 (T-cell immunoglobulin mucin 4), a transmembrane glycoprotein and member of the immunoglobulin superfamily, is exclusively expressed by antigen-presenting cells, which acts as the natural ligand of Tim-1, involved in T-cell expansion and cytokine production. Tim-4 is reported to be crucial for the phagocytosis of apoptotic cells by peritoneal macrophages, and studies have shown that interaction between Tim-1 on Th2 cells and Tim-4 on DCs, enhances Th2 cell function. The TIM-4 gene locus is associated with predisposition to asthma in both mice and humans and may be considered as a candidate disease gene for asthma. Studies have shown that Tim-4 significantly inhibits the efficient phagocytosis of apoptotic cells by PRMs (peritoneal resident Mac1+ cells), but not PEMs (peritoneal exudate Mac1+ cells) see Nakayama, M. et al. . for details.

Applications
Suitable for use in Immunofluorescence, Flow Cytometry and Immunohistochemistry. Other applications not tested.
Recommended Dilution
Immunohistochemistry: paraffin sections Optimal dilutions to be determined by the researcher.

Lymph node cells from immunized SD rats were fused with cells of the P3U1 myeloma cell line.

Storage and Stability
Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Note: Applications are based on unconjugated antibody.
Immunogen
Tim-4-Ig, consisting of aa1-288 from mouse Tim-4, coupled with the Fc portion of mouse IgG2a.

Supplied as a liquid in PBS, pH 7.2. Labeled with Fluorescein isothiocyanate (FITC).
Purity
Purified by Protein G affinity chromatography from tissue culture supernatant.
Specificity
Recognizes mouse TIM-4.
