Glutamic acid decarboxylase (GAD) catalyses the conversion of L-glutamic acid to the inhibitory neurotransmitter gamma-aminobutyric acid (GABA). Two forms of human GAD, GAD65 and GAD67, are encoded by two separate genes. Human GAD65 cDNA encodes a Mr 65,000 polypeptide, with 585aa residues, whereas human GAD67 encodes a Mr 67,000 polypeptide, with 594aa residues. GAD67 gene consists of 16 exons, spread over more than 45kb of genomic DNA. The GAD67 gene contains an additional exon (exon 0) that together with part of exon 1, specifies the 5' untranslated region of GAD67 mRNA. Human GAD67 shows 65% identity to GAD65 and is located in 2q31. GAD67 may play a role in the stiff man syndrome. Deficiency in this enzyme has been shown to lead to pyridoxine dependency with seizures.

Applications
Suitable for use in Western Blot. Other applications not tested.
Recommended Dilution
Western Blot: 0.1-0.5ug/ml Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile dH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Synthetic peptide corresponding to aa529-543, PQRREKLHKVAPKIK, from human GAD67 at C-terminus, different from the related mouse and rat sequences by 2aa.

Supplied as a lyophilized powder from PBS, 5% BSA, 0.05% thimerosal, 0.05% sodium azide. Reconstitute with 200ul sterile dH2O.

Purified by immunoaffinity chromatography.
Specificity
Recognizes human GAD67. Species Crossreactivity: mouse and rat
