Mouse Anti-AcCoA Carboxylase (Ser79), phosphorylated (MaxLight 650)

MaxLight™650 is a new Far-IR stable dye conjugate comparable to Alexa Fluor™647, DyLight™649, Cy5™ and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (655nm); Emission (676nm); Extinction Coefficient 250,000.

Acetyl-CoA carboxylase (ACC) is a complex multifunctional enzyme system. ACC is a biotin-containing enzyme which catalyzes the carboxylation of acetyl-CoA to malonyl-CoA, the rate-limiting step in fatty acid synthesis. There are two ACC forms, alpha and beta, encoded by two different genes. ACC-alpha is highly enriched in lipogenic tissues. The enzyme is under long term control at the transcriptional and translational levels and under short term regulation by the phosphorylation/dephosphorylation of targeted serine residues and by allosteric transformation by citrate or palmitoyl-CoA. The protein is localized to the cytoplasm in brain, placenta, skeletal muscle, renal, pancreatic and adipose tissues.
Applications
Suitable for use in Western Blot. Other applications not tested.
Recommended Dilutions
Western Blot: 0.5-2ug/ml. Detects phospho-Acetyl CoA Carboxylase (Ser79) in mouse heart cytosolic lysates. Mouse heart cytosolic lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with A0225-05A. Proteins were visualized using an IgG (HRP) goat anti-mouse and a chemiluminescence detection system. Optimal dilutions to be determined by the researcher.
Recommended Secondary Antibodies
I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt x Mo I1904-06H: IgG, H&L (HRP) (X-Adsorbed) Pab Gt x Mo
Recommended Positive Control
Acid Extracts from HeLa cells
Storage and Stability
Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™650 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.
Immunogen
Synthetic peptide containing (…SSM[pS]GL), in which [pS] corresponds to phospho-Serine at residue 79 of rat AcCoA Carboxylase conjugated to KLH.
Form
Supplied as a liquid in PBS, pH 7.2. No preservative added. Labeled with MaxLight™650.
Purity
Purified by Protein G affinity chromatography.
Specificity
Recognizes phosphorylated rat Acetyl CoA Carboxylase (Ser79), Mr 257kD. Species Crossreactivity: mouse.

Intended for research use only. Not for use in human, therapeutic, or diagnostic applications.
