Goat Anti-ADAMTSL-1 (Punctin Aggrecanase I, A Disintegrin And Metalloproteinase with ThromboSpondin-3 motif)
Produced in goats immunized with purified, NS0-derived, recombinant human Punctin (rhPunctin; aa29-525). Human Punctin specific IgG was purified by human Punctin affinity chromatography. Punctin is a secreted glycoprotein resembling members of the ADAMTS family although it lacks the propeptide region and the metalloproteinase and disintegrin-like domains. Punctin is expressed in adult skeletal muscle and may have important functions in the extracellular matrix. Alternative splicing of the Punctin transcript results in three isoforms. Isoform 1 is the longest and contains the entire isoform 2. Isoform 3 has the same sequence as isoforms 1 and 2 until residue 361, but a unique C-terminal end of 78 amino acids.
Applications
Suitable for use in ELISA, Western Blot, and Immunoprecipitation. Other applications not tested.
Recommended Dilution
ELISA: 0.5-1.0ug/ml. The detection limit for rhPunctin is approximately 1ng/well. Western Blot: 0.1-0.2ug/ml. The detection limit for rhPunctin is approximately 2ng/lane and 5ng/lane under non-reducing and reducing conditions, respectively. Immunoprecipitation: Used to immunoprecipitate rhPunctin from conditioned media of transfected NS0 cells. The recovered rhPunctin can be detected by western blot analysis using a monoclonal antibody. Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized powder may be stored at 4°C for short-term only. Reconstitute to nominal volume by adding sterile 40-50% glycerol and store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Form
Supplied as a lyophilized powder in 0.2um sterile-filtered solution in PBS with 5% trehalose. Reconstitute with sterile PBS. If 0.5ml of PBS is used, the antibody concentration will be 0.2mg/ml.
