As the key component of the cartilage extracellular matrix, aggrecan hydrates the collagen network and provides cartilage with its properties of compressibility and elasticity. Maintenance of aggrecan content is therefore critical to the function of the tissue and aggrecan degradation is an important factor in the erosion of articular cartilage in arthritic diseases. The deduced amino acid sequence of human aggrecan core protein consists of 2415 residues and predicts a signal peptide and domains of G1, IGD, G2, KS, CS1, CS2 and G3 (3). Two globular domains, G1 and G2, comprise the Nterminus of the proteoglycan and also contain link domains. The third globular domain, G3, corresponds to the Cterminus. The keratan sulfate (KS) and the chondroitin sulfate (CS) attachment domains are between G2 and G3. With KS and CS attached to the 250kD core protein, aggrecan monomers exist as a 1000-2000kD molecule.
Applications
Suitable for use in ELISA, Western Blot, Immunoprecipitation and Immunohistochemistry. Other applications not tested.
Recommended Dilution
Western Blot: 1ug/ml under non-reducing conditions only Immunohistochemistry: 8-25ug/ml in immersion paraffin-embedded human cartilage tissue sections. Immunoprecipitation: 25ug/ml immunoprecipitates recombinant human aggrecan from conditioned media of transfected NSO cells. Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recombinant protein corresponding to Val20-Gly675 from human Aggrecan G1-IGD-G2 Domains, expressed in NS0 cells.

Supplied as a lyophilized powder from PBS, 5% trehalose. Reconstitute with 1ml sterile PBS.
