Rabbit Anti-Aquaporin 5 (AQP5)

Water is a critical component of all living cells. Interestingly, tissue membranes show a great degree of water permeability. Mammalian red cells, renal proximal tubules, and descending thin limb of Henle are extraordinarily permeable to water. Water crosses hydrophobic plasma membranes either by simple diffusion or through a facilitative transport mechanism mediated by special protein “aquaporin”. Over the last decade, genes for several members of aquaporin family (AQP1-9 and AQPAP) have been cloned, expressed, and their distribution studied in many tissues. AQP5 (human, mouse, and rat 265aa) is found in several epithelial tissues (salivary and lachrymal glands, corneal epithelium in the eye and lung). It is implicated in generation of saliva, tears, and pulmonary secretions. AQP families of proteins are predicted to contain six transmembrane domains.

Applications
Suitable for use in ELISA, Western Blot, Immunohistochemistry and Immunofluorescence. Other applications not tested.
Recommended Dilutions
ELISA: 0.5-1ug/ml; Coat plates with control peptide at 1ug/ml. Western Blot: 1-10ug/ml (ECL) Immunohistochemistry (PFFE): 2-10ug/ml Immunofluorescence (PFFE): 2-10ug/ml Note: The use of 0.5-1% milk in all primary/secondary antibody-enzyme conjugate incubations is recommended to suppress non-specific bands. Optimal dilutions to be determined by the researcher.

Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Synthetic peptide corresponding to 17aa of rat Aquaporin 5, conjugatd to KLH. Epitope: C-terminal, Cytoplasmic domain. Species Sequence Homology: mouse, 100%; human, 80%; sheep, 76%.

Purity
Purified by immunoaffinity chromatography.
Specificity
Reconizes rat Aquaporin 5 (AQP5). No significant homology is seen with other AQPs or any other protein.
