MaxLight™650 is a new Far-IR stable dye conjugate comparable to Alexa Fluor™647, DyLight™649, Cy5™ and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (655nm); Emission (676nm); Extinction Coefficient 250,000
Bcl-X has 2 isoforms, Bcl-Xl (long) a 241 amino acid protein and Bcl-Xs (short) a 178 amino acid protein lacking a 63 amino acid domain that is well conserved among members of the Bcl-2 family.
Applications
Suitable for use in Flow Cytometry, Immunofluorescence, Immunoprecipiation and Immunohistochemistry. Not suitable for use in Western Blot. Other applications have not been tested.
Recommended Dilution
Immunoprecipitation (Use Protein A): 2ug/mg protein lysate Immunohistochemistry (formalin/paraffin): 2-4ug/ml for 30min at RT. Note: Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0 for 10-20min followed by cooling at RT for 20min. Optimal dilutions to be determined by the researcher.
Positive Control
Hodgkin’s lymphoma
Storage and Stability
Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™650 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.
Immunogen
Synthetic peptide corresponding to residues 3-14 (Cys-QSNRELVVDFLS) of human Bcl-X protein. Cellular Localization: Cytoplasmic and cell/nuclear membrane.
Form
Supplied as a liquid in PBS, pH 7.2. No preservative added. Labeled with MaxLight™650.
Purity
Purified by Protein A affinity chromatography.
Specificity
Recognizes Bcl-X at 27kD. Does not react with Bcl-2a or Bax protein. Species Crossreactivity: Human, mouse, rat and porcine.
