Mouse Anti-Bromodeoxyuridine (BrdU)

Bromodeoxyuridine (BrdU) is a thymidine analog and is specifically incorporated into DNA during DNA synthesis. Detection of bromodeoxyuridine (BrdU) incorporated into DNA is a powerful tool to study the cytokinetics of normal and neoplastic cells. In vitro or in vivo labeling of tumor cells with the thymidine analogue BrdU and the subsequent detection of incorporated BrdU with specific anti-BrdU monoclonal antibodies is an accurate and comprehensive method to quantitate the degree of DNA-synthesis. BrdU is incorporated into the newly synthezised DNA of the S-phase cells and can thus provide an estimate for the fraction of cells in S-phase. Also dynamic proliferative information (such as the S-phase transit rate and the potential doubling time) can be obtained, by means of bivariate BrdU/DNA flow cytometric analysis.

Applications
Suitable for use in ELISA and Immunocytochemistry. Other applications not tested.
Recommended Dilution
Immunocytochemistry: 1:3 dilution of this lot showed positive immunostaining for BrdU in HeLa cells fixed with 95% ethanol, 5% glacial acetic acid. Technical Tip: This antibody detects 5-Bromo-2- deoxyuridine incorporated into double-stranded DNA. DNA denaturation is not required. Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O containing 0.1% azide. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Form
Supplied as 4 vials. Each vial contains 2.5ul lyophilized mouse culture supernatant. Reconstitute each vial with 500ul ddH2O containing 0.1% azide.
