Mouse Anti-c-myc (Myc, myc-1, Cellular myelocytomatosis oncogene, Myc2, Niard, Nird, Transcription factor p64, v-myc avian myelocytomatosis viral oncogene homolog) (FITC)
The c-myc proto-oncogene is the cellular homologue v-myc, originally isolated from avian myelocytomatosis virus. c-myc protein is localized to the nucleus and exhibits cell cycle-dependent expression In human cell lines and neoplasms. Many types of abnormal c-myc expression have been observed including insertional inactivation, translocation, and amplification. c-myc is involved in the control of cell proliferation and differentiation. Translocation of the c-myc locus on chromosome 8 to the immunoglobulin loci on chromosome 14 (heavy chain); 2 (k light chain); or 22 (l light chain) is described in Burkitt’s lymphoma and other B-cell lympho-proliferative conditions. An aberrant expression of the c-myc gene occurs in tumors of different origins such as colorectal, gastric, gallbladder, hepatic, mammary, ovarian, endometrial, head and neck, pulmonary, prostatic, thyroidal, oral, ocular, nasopharyngeal, endocrine, as well as hematopoietic neoplasms.
In addition, deregulated expression of c-Myc induces apoptosis in different cell types, with c-myc requiring p53 for apoptosis in many cell types. This fact indicates heterogeneous mechanisms for c-myc-induced apoptosis.
Applications
Suitable for use in Flow Cytometry. Other applications not tested.
Recommended Dilutions
Flow Cytometry: Neat, 10ul labels 10e6 cells in 100ul. Membrane permeabilization is required. Optimal working dilutions to be determined by researcher.
Hybridoma
Sp2/0 myeloma cells with spleen cells from Balb/c mice.
Storage and Stability
Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Synthetic peptide corresponding to aa408-439 at the C-terminal region of human c-myc, conjugated to KLH.
Form
Supplied as a liquid in PBS, pH 7.2, 1% BSA, 0.09% sodium azide. Labeled with Fluorescein isothiocyanate (FITC).
Purity
Purified by Protein G affinity chromatography.
Specificity
Recognizes the sequence EQKLISEEDL of human c-Myc and may be used to detect proteins and peptides labeled with molecular tags containing this sequence.
