Rat Anti-CCR5 (C-C Chemokine Receptor Type 5, CCCKR5, CC-CKR-5, C-C CKR-5, CKR5, IDDM22, CCR-5, AM4-7, CD195, CHEMR13, CMKBR5, HIV-1 Fusion Coreceptor)

CCR5 (CC chemokine receptor 5; also CD195 and MIP-1a receptor) is a 41-44kD member of the G-protein coupled receptor #1 family of proteins. It is expressed on Treg cells, NK cells, neurons, macrophages and Th1 cells. CCR5 mediates cell adhesion and migration induced by several chemokines including CCL3/MIP-1 alpha, CCL4/MIP-1 beta, CCL5/RANTES, and CCL8/MCP-2. It also functions as a coreceptor for macrophage-tropic HIV-1 infection. CCR5 contains an O-glycosylated and sulfated extracellular N-terminus (aa1-32), and a phosphorylated and palmitoylated intracellular C-terminus (aa304-354). CCR5 will form homodimers, heterodimers with CCR2, and heterooligomers with CCR2 and CXCR4. Within aa1-32, mouse CCR5 shares 72% and 91% amino acid sequence identity with human and rat CCR5, respectively.

Applications
Suitable for use in ELISA and Western Blot. Other applications not tested.
Recommended Dilutions
Western Blot: Using RAW 264.7 mouse monocyte/macrophage cell line treated with LPS under non-reducing conditions only Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recombinant protein corresponding to Met1-Ala32 from mouse CCR5 expressed in E. coli.
Form
Supplied as a lyophilized powder from PBS, pH 7.4, 5% trehalose. Reconstitute with 200ul sterile PBS.

Purified by Protein G affinity chromatography from hybridoma culture supernatant
Specificity
Recognizes mouse CCR5. Does not crossreact with recombinant mouse CCR1, 4 or 8.
