Hamster Anti-CD11c (Monocyte, Macrophage, Integrin alphaX) (FITC)

CD11c antigen is expressed mainly on monocytes and in low density on granulocytes and NK cells. High levels of CD11c are expressed on tissue macrophages. It is also found on lymphoid cell lines including hairy cell leukemia. CD11c (integrin aX subunit) combines with CD18 (integrin b2 subunit) to form the integrin protein 150,95 (aXb2, CD11c/CD18). CD11c/CD18 has been reported to play an important role in cytotoxic T cell killing, and in neutrophil and monocyte adhesion to endothelium. Although its ligands in these two cases have not been identified. CD11c/CD18 is not expressed on leukocytes of patients with leukocyte adhesion deficiency. leukocyte adhesion deficiency results from defects in the CD18 gene, leading to diminished expression of all CD18-containing integrins. CD11c antibodies bind multiple ligands including the complement fragment iC3b, CD54 (ICAM-1), fibrinogen and bacterial lippolysaccharide.

Applications
Suitable for use in Flow Cytometry, Immunoprecipitation and Immunohistochemistry. Other applications not tested.
Recommended Dilution
Flow Cytometry: 1:100. Use 10ul of the suggested working dilution to label 1x10e6 cells in 100ul. Optimal dilutions to be determined by the researcher.
Hybridoma
Spleen cells from immunized Armenian Hamster were fused with cells of the Sp2/0 myeloma cell line.

Storage and Stability
May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. FITC conjugates are sensitive to light.

Mouse spleen dendritic cells.

Supplied as a liquid in PBS, pH 7.4, 1% BSA, 0.09% sodium azide. Labeled with fluorescein isothiocyanate (FITC).
Purity
Purified by Protein G affinity chromatography.
Specificity
Recognizes the murine homologue of human CD11c. Has been reported to enhance antigen specific responses when used to target dendritic cells in vivo (3).
