Mouse Anti-CD14 (CD14 Antigen, Lipopolysaccharide Receptor, LPSR, Monocyte Differentiation Antigen 14, Myeloid Cell Specific Leucine Rich Glycoprotein) (FITC)
CD14 antigen, Lipopolysaccharide receptor, LPSR, Monocyte Differentiation Antigen 14, Myeloid cell specific leucine rich glycoprotein
Custered as porcine CD14 at the Third International Workshop on Swine Leukocyte Differentiation Antigens (Haverson et al. 2001). Immunoprecipitates a protein of ~50kDa consistent with the expected apparent molecular weight of porcine CD14, and demonstrates the expected CD14 profile by dual labelling and competition studies. Further, pre-incubation of peripheral blood monocytes with MIL2 inhibits the binding of FITC labelled LPS, consistent with masking the CD14 LPS binding site (Thacker et al. 2001).
Clone MIL2 demonstrates staining of both monocytes and neutrophils in peripheral blood by flow cytometry. Lymphocytes and eosinophils are negative for MIL2 staining (Zelnickova et al. 2007). Cloning and characterization of porcine CD14 indicates a high degree of both functional and structural conservation when compared to CD14 from other mammalian species, the gene maps to chromosome 2 and is expressed on a wide range of tissues in a manner consistent with expression on myeloid cells. (Petersen et al.2007 and Sanz et al. 2007).
Applications
Suitable for use in Flow Cytometry. Other applications not tested.
Recommended Dilution
Flow Cytometry: 10ul labels 10e6 cells in 100ul Optimal dilutions to be determined by the researcher.
Hybridoma
P2-X63-Ag.653 myeloma cells with spleen cells from Balb/c mice.
Storage and Stability
Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Porcine peripheral blood lymphocytes.
Form
Supplied as liquid in PBS, 1% BSA, 0.09% sodium azide. Labeled with fluorescein isothiocyanate (FITC).
Purity
Purified by Protein G affinity chromatography from tissue culture supernatant.
Specificity
Recognizes porcine CD14. Species crossreactivity: human
