Mouse Anti-CD16 (Fc gamma RIII alpha, NK cells) (Biotin)

Human CD16 is a low affinity receptor for complexed IgG and occurs in two distinct forms, as a glycosyl phosphtidyl inositol (GPI) anchored protein in granulocytes, and as an integral membrane protein on NK cells and macrophages.

Applications
Suitable for use in Flow Cytometry. Other applications not tested.
Recommended Dilutions
Flow Cytometry: 5ug/ml labels 5x10e5 ficoll prepared human peripheral blood mononuclear cells Optimal dilutions to be determined by the researcher.
Flow Cytometry Protocol
1. Five x 10e5 ficoll prepared human peripheral blood mononuclear cells were washed and incubated 45 minutes on ice with 80ml of C2267-33B at 5ug/ml. 2. Cells were washed twice and incubated with S7973-88P: Streptavidin (PE) 3. Washed cells three times, fix and analyzed by FACS. 4. A net 19% subpopulation of the cells stained positive with a mean shift of 2.4 log10 fluorescent units when compared to I1904-76K: IgG1 Murine Negative Control (Biotin) at a similar concentration. 5. Binding was blocked when cells were pre-incubated 10 minutes with 20ul of 0.5mg/ml C2267-33A: CD16 (Fc gamma RIII alpha, NK cells).
Storage and Stability
Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Purified by Protein A affinity chromatography.
Specificity
Recognizes both allelic forms of human CD16. Blocks binding of complexed IgG to CD16.
