Mouse Anti-CD54 (Intercellular Adhesion Molecule, ICAM-1) (PE)

Human CD54 (ICAM-1) mediates cell adhesion by binding to the integrins CD11a/CD18 (LFA-1) and to CD11b/CD18 (Mac-1). CD54 expression on resting peripheral blood leukocytes in weak but is upregulated on activated T and B lymphocytes and on monocytes.

Applications
Suitable for use in FLISA, Flow Cytometry, Immunohistochemistry and Western Blot. Other applications not tested.
Recommended Dilution
Flow Cytometry: 1:50 Immunohistochemistry: Frozen sections Optimal dilutions to be determined by the researcher.
Flow Cytometry Protocol
1. Five x 10e5 cultured human Daudi cells were washed and preincubated ~5 minutes with 20ul of 0.25mg/ml human IgG (to block non specific binding) after which they were incubated 45 min on ice with 80ul of C2409-10D at a dilution of 1:50. 2. Cells were washed three times, fixed and analyzed by FACS. 3. Cells stained positive with a mean shift of 1.78 log10 fluorescent units when compared to a Mouse IgG1/R-PE negative control at a similar concentration. 4. Binding was 99% blocked when preincubated 10 min with 20ul of C2409-10A at a concentration of 0.5mg/ml.
Storage and Stability
May be stored at 4°C before opening. DO NOT FREEZE! Stable at 4°C as an undiluted liquid. Dilute only prior to immediate use. Stable for at least 12 months at 4°C. Freezing R-Phycoerythrin (PE) conjugates will result in a substantial loss of activity. PE conjugates are sensitive to light.

Specificity
Recognizes the D1 domain of the human CD54 molecule of 90kD. Inhibits CD54 binding to LFA-1.
