Applications
Suitable for use in Immunofluorescence, Immunocytochemistry, RIA, ELISA and Inhibition of BoTox-B binding to Neuro2A Cells. Other applications not tested.
Recommended Dilutions
Immunofluorescence (IC): 1:10 stains Neuro2A cells treated with GT1b ganglioside prior to the addition of the toxin (BoTox-B) using an indirect staining procedure. Following treatment, the cells were washed 4 times, fixed in a neutral buffered formalin for 10 minutes, permeablized and blocked. Reacted with C5851-48B for 60 minutes and then probed with a secondary antibody; IgG (FITC) Pab Gt x Mo. Epifluorescent microscopy was used to observe staining pattern. Inhibition of BoTox-B binding to Neuro2A Cells: Inhibits the binding of BoTox-B to Neuro2A cells treated with GT1b ganglioside prior to the addition of the toxin, and prior incubation of C5851-48B with the synthetic peptide immunogen has been shown to eliminate the inhibition of cellular binding. Optimal dilutions to be determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Synthetic peptide analog of the carboxyl terminal region of BoTox-B.

Specificity
Recognizes Clostridium botulinum B Toxoid (BoTox-B) (1278-1291). Binds to intact whole BoTox-B by ELISA, RIA and cell staining techniques. Does not react with BoTox-A, BoTox-C, BoTox-E and BoTox-F (1272-1280).
