The cohesin complex plays an important role in tethering sister chromatids during the prophase through anaphase stages of mitosis, making certain that genomic information is replicated accurately. As the cellular division process continues, separase destroys the cohesin complex by means of cleavaging its Scc1 subunit (Mcd1/Scc1/hRad21), allowing the chromatids to separate and divide with the cell. During apoptosis the C-terminal portion of hRad21 is cleaved and translocated to the cytoplasm. This cleavage and translocation may act as a nuclear signal that promotes and accelerates subsequent events of apoptosis (2).

Suitable for use in Western Blot. Other applications not tested.
Recommended Dilution
Optimal dilutions to be determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot Store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
An N-terminal fusion protein of the C-terminal fragment of Scc1 subunit human cohesin (amino acids 544-631 of hRad21).
Form
Supplied as a liquid in PBS, pH 7.2.
Specificity
Specific for both the nuclear hRad21 protein and the cleaved cytoplasmic hRad21 protein. Species Crossreactivity: human and mouse cohesin (Rad21) protein. Other species have not been tested.
