Goat Anti-Complement C1rLP (C1r Like Protein, CLSPa)
Mouse C1rLP, also CLSPa, is a 55kD, secreted, atypical glycoprotein member of the AGY group of servine proteases. It has protease activity for pro-haptoglobin, but accomplishes cleavage without a typical Asp in the catalytic site. In addition, it is proposed to block unwanted complement-mediated proteolysis. Mature C1rLP is 443 amino acids (aa) in length. It contains one CUB domain (aa # 43-165), a truncated CCP2 region (aa # 167-223) and a serine protease domain (aa # 239-482). There is a potential short form that utilizes an alternate start site at Met 27. Over aa's # 44-456, mouse C1rLP is 78% and 90% aa identical to human and rat C1rLP, respectively.
Applications
Western Blot: This antibody can be used at 0.1-0.2ug/ml with the appropriate secondary reagents to detect mouse C1rLP. The detection limit for rmC1rLP is approximately 2 ng/lane under non-reducing and reducing conditions. Immunoprecipitation: This antibody has been used to immunoprecipitate rmC1rLP from conditioned media of transfected NS0 cells. Direct ELISA: This antibody can be used at 0.5-1.0ug/ml with the appropriate secondary reagents to detect mouse C1rLP. The detection limit for rmC1rLP is approximately 0.07 ng/well. Optimal dilutions should be determined by each laboratory for each application.
Storage and Stability
Lyophilized powder may be stored at 4°C for short-term only. Reconstitute to nominal volume by adding sterile PBS with 40-50% glycerol and store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
NS0-derived rmC1rLP (aa18-456)
Form
Supplied as a lyophilized powder from PBS, 5% trehalose. Reconstitute with 500ul sterile PBS.
Purity
Purified by immunoaffinity chromatography.
Specificity
Selected for its ability to recognize mouse C1rLP in direct ELISAs and Western Blots. In these formats, this antibody shows approximately 40% crossreactivity with rhC1rLP and less than 1% crossreactivity with rhC1R and rhC1S.
