Mouse Anti-Desmoplakin 1,2

Desmoplakin (DP) is a cytoplasmic plaque protein that is located in desmosomes, which are abundant in tissues subjected to mechanical stress, like muscle and epidermis. The role of DP is to link the transmembrane cadherins via plakoglobin to the cytoplasmic intermediate filament network. DP comprises an N-terminal, desmosome-associated plakin domain binding to plakoglobin and plakophilin, a central coiled-coil rod domain responsible for dimerization, and a C-terminal domain consisting of three homologous plakin repeat subdomains, named “A”, “B”, and “C”, that interact with the intermediate filaments. Alternative splicing generates two isoforms (DP I and DP II) that differ in their rod domain lengths, with relative molecular weights of 332kD and 259kD, respectively.

Applications
Suitable for use in Western Blot, Immunohistochemistry and Immunofluorescence. Other applications have not been tested.
Recommended Dilutions
Immunohistochemistry (Frozen): 1:10 Optimal dilutions to be determined by the researcher.
Recommended Positive Controls
Human - A431 & MCF-7; Rat - MH1C1; Bovine - MDBK & BMGE.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Supplied as a lyophilized powder from PBS, pH 7.4, 0.5% BSA, 0.09% sodium azide. Reconstitute with 1ml sterile ddH2O.
Purity
Purified from culture supernatant.
Specificity
Recognizes bovine desmoplakins 1 and 2 from stratified epithelia and simple epithelia, including glands, urothelium, thymic reticular epithelium, hepatocytes, intercalated disks of myocardium and arachnoid cells of meninges. Shows distinct punctate membrane staining of desmoplakins 1 (250kD) and 2 (215kD). |Species Crossreactivity: human, rat, mouse and chicken.
